Fas ligand (FasL or CD95L) is a type-II transmembrane protein that belongs to the tumor necrosis factor (TNF) family. Fas ligand/receptor interactions play an important role in the regulation of the immune system and the progression of cancer. It is mapped to 1q24.3. FASL may contribute to the immune privilege of tumors. Pharmacologic products that render infiltrating T cells insensitive to FASL-induced killing may break the immunologic unresponsiveness to melanoma and provide a complementary approach in the therapy of malignant melanoma. FASL-mediated apoptosis is important for skin homeostasis, the dysregulation of FAS-FASL interactions may be central to the development of skin cancer. |
